What triggers (early) astrocyte reactivity?

Abstract:Microglia respond rapidly to brain injury, yet the signaling pathways that determine whether these responses are protective or detrimental remain incompletely understood. In this talk, I will discuss recent work identifying the Trem2–Gpnmb axis as a key regulator of microglial function across distinct neurological disorders. Using a photothrombotic stroke model, we found that Trem2 exacerbates ischemic brain injury through Gpnmb-dependent mechanisms, linking microglial activation to worsened tissue damage and neurological outcome. I will then extend this framework to Alzheimer's disease, where our work suggests that Gpnmb is not merely a marker of disease-associated microglia, but an active modulator of microglial phenotype and pathological progression. By comparing stroke and Alzheimer's disease, this presentation will highlight how the Trem2–Gpnmb pathway may represent a shared molecular program through which microglia interpret injury and neurodegeneration.
